Somatotropin Penetration Testing from Formulations Applied Topically to the Skin

چکیده

Growth hormone (somatotropin—STH) deficiency therapy requires daily injections of recombinant human growth hormone. The FDA approved treatment with STH one dose per week for the first time in 2021. However, injectable drug application is accompanied by numerous inconveniences. Therefore, an attempt was made to formulate a less invasive formulation topical skin. A substrate prepared based on polymer, methylcellulose (MC), into which introduced at concentration 1 mg/g. Simultaneously, formulations were STH, albumin (ALB) added different concentrations: 0.1%, 0.2% and 0.5%. test degree permeation carried out, as well effect ALB parameters. Selected rheological properties obtained investigated. simulated vivo conditions through porcine skin indicated relatively good bioavailability over 80% confirmed effectiveness MC carrier prolonged penetration rate increased 93%. hydrogels found be typical shear-thinning, thixotropic fluids.

Topically applied ceramide accumulates in skin glyphs

Ceramides (CERs), structural components of the stratum corneum (SC), impart essential barrier properties to this thin outer layer of the epidermis. Variations in CER species within this layer have been linked to several skin diseases. A recent proliferation of CER-containing topical skin-care products warrants the elucidation of CER penetration profiles in both healthy and diseased skin. In the...

Follicular penetration of topically applied caffeine via a shampoo formulation.

AIMS Follicular drug delivery is the prerequisite for an effective treatment of androgenetic alopecia or other reasons of premature hair loss. METHODS The follicular penetration of caffeine, applied topically in a shampoo formulation for 2 min, was measured with highly sensitive surface ionization in combination with mass spectroscopy, a selective method for the detection of very small quanti...

Ocular absorption of topically applied FK506 from liposomal and oil formulations in the rabbit eye.

PURPOSE To investigate the use of topically applied FK506, a new immunosuppressive compound, systemic and ocular absorption was determined in serum and various ocular tissues. METHODS Two drops of 20 microliters FK506 were applied using oil dissolved (OD-FK506) or liposome-bound (LIP-FK506) drug. FK506 concentrations were measured at intervals of 30, 60, and 120 minutes by immunoassay. RESU...

Systemic toxicity from topically applied salicylic acid.

Topically applied salicylic acid has enjoyed long and extensive usage as an agent to enhance desquamation in various hyperkeratotic skin diseases. In some of these disorders much of the skin surface may be affected, resulting in the potential for significant percutaneous absorption of the drug. We describe a patient who gradually developed evidence of severe salicylate toxicity while being trea...

Contribution of the Hair Follicular Pathway to Total Skin Permeation of Topically Applied and Exposed Chemicals

Generally, the blood and skin concentration profiles and steady-state skin concentration of topically applied or exposed chemicals can be calculated from the in vitro skin permeation profile. However, these calculation methods are particularly applicable to chemicals for which the main pathway is via the stratum corneum. If the contribution of hair follicles to the total skin permeation of chem...
